Project description:
A range of organ and cellular model systems (including liver, breast, colon, adipocytes) will be investigated with innovative human-relevant NAMs, including state of the art transcriptomic approaches, high-content analysis, cell painting, epigenetics and high-throughput-compatible reporter systems addressing well established and relevant mechanisms of carcinogenesis, such as (oxidative) stress, epithelial-mesenchymal transition (EMT), inflammation, changes in metabolism, epigenetic marks or nuclear receptors signaling as well as proliferation. In addition, experimental approaches will be complemented by the optimization of in silico tools for the identification of NGTxCs to address the assay needs for the development of an NGTxC IATA in cooperation with WP6 and ongoing activities at EU and OECD level. The project will improve our knowledge of the relationships between a range of different substances specific for distinct adverse outcome pathways that ultimately lead to cancer. In addition, methods with increasing complexity, from 2D cell culture up to 3D spheroids and zebrafish as a simple vertebrate model system, will facilitate the integration of the physiological and toxicological relevant information with respect to adversity in order to inform about potential carcinogens and to improve chemical assessment and regulation. The project will align with AOPs and IATA work under Task 5.3 as well as WP6 and takes into account the ongoing work of the OECD expert group on non-genotoxic carcinogens as well as current EU initiatives.
